mRNA vaccines, basic technology for controlling adverse effects

Dr. Satoshi Uchida and his team, belonging to R&D Theme 2 at Tokyo Medical and Dental University, have developed a new technology to reduce the side effects of messenger RNA (mRNA) vaccines. This technology delivers mRNA directly to immune cells in the skin without using particles that carry the mRNA, thereby reducing side effects such as arm pain and fever. Conventional mRNA vaccines have become widespread for COVID-19, but side effects have been reported following administration. These side effects are thought to be caused by the particles containing the mRNA.

The new technology utilizes a needle-free injector that delivers mRNA into the body instantly using the force of a small amount of gunpowder. Experiments in monkeys and mice confirmed that the mRNA reaches immune cells before it can be broken down, resulting in lower levels of inflammation compared to traditional vaccines. As a result, almost no side effects were observed, and antibodies were generated within the animals, confirming the vaccine's function. This technology has the potential to contribute to safer vaccine administration.
